Detection of Kudoa septempunctata in the Japanese Flounder by LoopMediated Isothermal Amplification (LAMP) Targeting the Internal Transcribed Spacer (ITS) Regions

DOI : 10.4194/1303-2712-v16_3_15 Viewed : 3913 - Downloaded : 3498 Recent food poisoning outbreaks from consumption of the Japanese flounder, Paralichthys olivaceus infested with a myxozoan parasite, Kudoa septempunctata have become a serious human health concern. In this study, we sequenced the internal transcribed spacer (ITS) regions between the 18S and 28S rRNA genes of K. septempunctata from cultured flounder in Japan and imported flounder from Korea. Sequence analysis indicates that the ITS1 regions are highly conserved in Japanese and Korean isolates of the parasite. A loop-mediated isothermal amplification (LAMP) assay targeting the ITS1 region was developed for rapid, sensitive and specific detection of either isolate of the parasite.
